Two possible metabolic consequences of the different levels of cellular function that were induced in rats by retinol, retinoic acid and 7,8-benzoflavone were investigated: (1) whether there was an inhibition of carcinogenesis by retinol, retinoic acid or 7,8-benzoflavone when liver cells in the early stages of carcinogenesis were the only functioning cells in the body, and (2) whether there was a worsening of the effects of these chemicals on carcinogenesis when intact animals were employed and liver cells were the first to have a metabolic effect. Under these conditions there was no inhibition of hepatocarcinogenesis by dietary retinol or the retinoid mixture. When 7,8-benzoflavone was given in the drinking water, however, it did block hepatocarcinogenesis. If the above regimen was begun immediately after carcinogen treatment, there was a slightly greater inhibition of carcinogenesis by 7,8-benzoflavone than if the dose was begun 24 hr after carcinogen treatment.
